Density gradient centrifugation ( formally known as the percoll gradient )
1.A density gradient is made from silica-coated particles. Bottom layer 80%, top layer 40%. Semen is placed on top. |
2.Centrifugation is used to separate the immotile debris from the motile spermatozoa. Motile spermatozoa are filtered out and accumulate at the bottom. |

3.The sample is washed in culture media. |
4.The most motile spermatozoa are counted and analyzed and used for I.U.I., insemination or ICSI. |
Swim up
1.Test tube with culture media is placed at an angle of 45o. |
2. After a period of time motile spermatozoa is aspirated from the top portion of the media after swimming up from the semen. | ||
3. The most motile spermatozoa are counted and analyzed and used for insemination. |
