How spermatozoa is prepared

Sperm recovery -How spermatozoa is prepared for IUI & IVF

Density gradient centrifugation ( formally known as the percoll gradient )

1.A density gradient is made from silica-coated particles.

Bottom layer 80%, top layer 40%.

Semen is placed on top.

2.Centrifugation is used to separate the immotile debris from the motile spermatozoa.

Motile spermatozoa are filtered out and accumulate at the bottom.

3.The sample is washed in culture media.

Swim up

 

1.Test tube with culture media is placed at an angle of 45o.

 
Semen is underlayed at the bottom.

3.  The most motile spermatozoa are counted and analyzed and used for insemination. sperm
